Malaria Vaccine Candidate Produces Disappointing Results in Clinical Trial

The latest clinical trial of the world?s leading malaria vaccine candidate produced disappointing results on Friday. The infants it was given to had only about a third fewer infections than a control group.

But researchers said they wanted to press on, assuming they keep getting financial support, because the number of children who die of malaria is so great that even an inefficient vaccine can save thousands of lives.

Three shots of the vaccine, known as RTS, S or Mosquirix and produced by GlaxoSmithKline, gave babies fewer than 12 weeks old 31 percent protection against detectable malaria and 37 percent protection against severe malaria, according to an announcement by the company at a vaccines conference in Cape Town.

Last year, in a trial in children up to 17 months old, the same vaccine gave 55 percent protection against detectable malaria and 47 percent against severe malaria.

The new trial ?is less than we?d hoped for,? Moncef Slaoui, chairman of research and development at Glaxo, said in a telephone interview. ?But if a million babies were vaccinated, we would prevent 260,000 cases of malaria a year. This is a disease that kills 655,000 babies a year ? 31 percent of that is a very large number.?

The company, which has already spent more than $300 million on the vaccine, wants to keep forging ahead, Mr. Slaoui said, ?but it is not just our decision.?

It also depends on the PATH Malaria Vaccine Initiative, which has put more than $200 million of its Bill and Melinda Gates Foundation financing into the vaccine, and on the World Health Organization, which has helped talk seven African countries into allowing the vaccine to be tested on their children.

The Gates Foundation declined to say how much money it was ultimately prepared to spend on an imperfect vaccine; this set of trials is set to go into 2014.

?The efficacy came back lower than we had hoped, but developing a vaccine against a parasite is a very hard thing to do,? Bill Gates said in a prepared statement. ?The trial is continuing, and we look forward to getting more data to help determine whether and how to deploy this vaccine.?

All the families in the trial were given insecticide-treated mosquito nets and encouraged to use them; 86 percent did, so the vaccine?s results were achieved on top of other anti-malaria measures.

RTS, S contains a protein found on the parasite?s surface that provokes an immune reaction. It was first identified decades ago by two New York University scientists, Ruth and Victor Nussenzweig. The vaccine was developed by Glaxo in Belgium and initially tested on American volunteers by the Walter Reed Army Institute of Research.

When the Gates Foundation began focusing on global health in the early part of this century, it was one of the first projects the foundation adopted. Different ways to make the vaccine more effective, including adding different boosters and giving more shots, are being experimented with. Other vaccines using different ways to provoke an immune reaction exist, but none are as far along in clinical trials.

Like an H.I.V. vaccine, one against malaria has proved an elusive goal. The parasite morphs several times, exhibiting different surface proteins as it goes from mosquito saliva into blood and then into and out of the liver. Also, even the best natural ?vaccine? ? catching the disease itself ? is not very effective. While one bout of measles immunizes a child for life, it usually takes several bouts of malaria to confer even partial immunity. Pregnancy can cause women to stop being immune, and immunity can fade out if someone moves away from a malarial area ? presumably because they no longer get ?boosters? from repeated mosquito bites.

Google kicks Xperia S Android Open Source Project out of its nest, Sony takes it under its wing (video)

After just under three months of development, Google's wrapped up its experimental work on bringing the Sony Xperia S into the Android Open Source Project fold. According to Sony, AOSP Technical Lead Jean-Baptiste Quéru considers the effort a success, but the device is being taken off the project's roadmap so Mountain View can focus on its own hardware. Currently, an AOSP build boots on the Xperia S hardware with support for SD-Cards, Wi-Fi and its built-in sensors. Audio and the phone's modem are also operational, but they require proprietary binaries Hirai and Co. can't publish just yet. Work on polishing the handset's vanilla Android experience isn't over, however. Sony has moved the code to its GitHub account and is welcoming developers to pitch in and help with the open source effort. DIY Multiple Roll Tape DispenserIn your home workshop or craft area you probably use a few types of tape depending on your work. It might be handy to have one jumbo dispenser that handles all types of tape that you regularly use. Here's how to make one out of scrap lumber, a dowel, and a hacksaw blade.

Woodworking weblog American Woodworker needed such a dispenser and made one from ?-inch lumber to build boxes to hold various types of tape. In the side panels small cuts were made so that a filler block can be inserted into the standard three-inch diameter opening in a tape roll and replaced as needed. A hacksaw blade is mounted to the edge of the dispenser for easy cutting. The filler blocks are made from 2-3/16-inch blocks with rounded corners and a enough of a ?-inch dowel to fill the gaps in the side panels.

LIPA Communication Leaves Many in the Dark - Three Village Patch

So far, the only information Colette Adamo Grosso said she received from the Long Island Power Authority was a recorded phone message two days before the hurricane hit last week, letting her know she could be without power for seven to 10 days.

"My best intel has come from Facebook news feed from neighbors, and community groups. Next from elected officials, who are obviously working hard to get help to all in need and communicate the information," she said in a message to Miller Place-Rocky Point Patch via Facebook. "Last is LIPA."

Grosso is not alone. While elected officials like State Sen. Ken LaValle, R-Port Jefferson, and Suffolk Legis. Kara Hahn, D-Port Jefferson, have been making frequent updates on the outage situation via social media and email following municipal conference calls with LIPA, some locals say the utility company has left them in the dark in more ways than one.

"I get updates from news, or friends ... but have not heard from LIPA since this mess started last week," Laurie Hansen said via the Three Village Patch Facebook page.

In Port Jefferson ? where LIPA officials have said repairs are more complicated and complete fixes may take longer than initially thought ? Mayor Margot Garant said she has received "nothing but complaints" from residents regarding LIPA's communication with them.

"They update us two times a day with a conference call, which is very basic," she said in an email to Patch. "But there is no direct connection to the people. They are frustrated."

Brookhaven councilwoman Jane Bonner, who said she has been relaying information to her residents via email, telephone, and even face-to-face visits, agreed with Garant.

"People feel like they?re in the dark. They don?t feel like they have any communication with LIPA," Bonner said. "They have no electricity, no internet. There?s no cell phone service. LIPA wants them to call the 1-800 number, and they don?t have the telephone to call with."

Hahn said LIPA needs to take a multi-pronged approach to communication with its customers. She herself signed up for LIPA's text message alerts, but it was a lengthy process to get it to work properly, and the alerts were few and far between.

"Once they started, I should have gotten one of those every day telling me something. 'We know you?re still out,'" she said. "I feel like they should have used it more. But I understand that this was a bad storm. Everybody?s very frustrated."

Barbara Schindler Geraci was among those frustrated by the text message system. "Signed up for text messages, but the updates have been generic and of no use to me," she said via Facebook.

Another Facebook commenter, Karla Maroney, said she chose email updates from LIPA rather than text messages. "I get several updates a day," she said.

Still another, Jenney Dzwilewski, said: "I don't get updates ? only what is said on the radio! You get disconnected everytime you call. ... It's so unreal!"

LIPA officials could not be reached for comment.

A LaValle representative said that "all of these issues are going to be addressed after power is restored to all Long Islanders."

At least one local official, Poquott?Village Mayor Barbara Donovan, has said LIPA's communication with the municipalities was all that was needed in their village.

"It worked very well because I have very good communication with the residents. They were able to come into village hall and ask, or call," Donovan said. "We couldn?t tell them anything more than LIPA told us. But they knew we were getting an update at 11 a.m. and at 4:30 p.m., so some of them would come in at noon or 5:30 and ask."

Deep South Dish: Ground Beef Hobo Stew

Pantry friendly and adaptable, I just love this stew. In some variation and often depending on the type of meat you use, you may know this dish by the names of Campfire Stew, Girl Scout Camp Stew, Hobo Stew, Ground Beef Stew, Beggar Stew, Mulligan Stew, or V8 Stew - just to name a few. I just settled on calling it a Ground Beef Hobo Stew.

Historically, a basic Dutch oven style stew that dates back to the early 1900s, it was slow cooked over the coals of an open campfire, often in a can (remember that from Girl Scouts?), or made up in a large casserole style foil packet. It was a communal dish, often enhanced by the next contributor who showed up through the woods, with whatever he had. We'll just keep it easy and take it stovetop or crockpot here.

There really aren't any hard and fast rules as to what goes in it, except some kind of meat, potatoes, a mixture of veggies and not much liquid. What else you add in, depends on what you have on hand and that is why I love it so much. It is a stew though, not a soup, so it should be thick and hearty, although with more liquid added, it also makes a mighty fine soup.

For my stew, I like to use V-8 as my primary liquid. I actually like and used to drink V-8 regularly, and no, not only in a Bloody Mary, though I certainly have enjoyed one of those a time or two in my life. I used to add it to soups and stews where I used tomatoes,? though I'm not really quite sure why I stopped, except that V-8 got a little more expensive than my budget allowed, and it wasn't really a pantry staple for me like canned tomatoes are. The blend of veggies in tomato juice found in V-8 is a perfect accent though for just about any soup or stew where you normally use tomatoes, so it's really no secret that cook's have been enhancing recipes with it for years.

This really is a wonderful stew, thick, hearty, filling - just perfect for these chilly days, and very easily adaptable to the slow cooker. While not everybody does, I prefer to brown and drain ground meat before adding it to a crockpot recipe, but other than that, it's just dump all the ingredients in and let it cook till the veggies are tender.

Recipe: Ground Beef Hobo Stew

?From the Kitchen of Deep South Dish
Prep time: 10 min |Cook time: 2 hours | Yield: About 6 to 8 servings

Ingredients
  • 1 tablespoon of oil
  • 1 cup of chopped onion
  • 1/2 cup of chopped mixed sweet and hot peppers
  • 1/2 cup of celery
  • 1 tablespoon of minced garlic
  • 1-1/2 pounds of ground beef
  • 2 tablespoons of all purpose flour
  • 1-1/2 cups of beef stock or broth
  • 3 (11.5 ounce) cans of original or spicy V-8, or tomato juice
  • 1 (14.5 ounce) can of diced tomatoes
  • 2 large potatoes, peeled and diced
  • 2 cups of fresh diced carrots (about 2 large)
  • 2 cups of corn
  • 2 cups of cut green beans
  • Dash of Worcestershire sauce
  • Couple dashes of hot sauce
  • 1/4 teaspoon of Cajun seasoning (like Slap Ya Mama), or to taste, optional
  • 1 teaspoon of kosher salt, or to taste
  • 1/4 teaspoon of freshly cracked black pepper, or to taste
Instructions

In the bottom of a Dutch oven or other heavy pot, heat the oil over medium high heat. Add the onion, peppers and celery; cook and stir until softened, about 4 minutes. Add the garlic and ground beef, cook and stir until meat is browned and cooked through. Drain off excess fat. Sprinkle meat with flour; cook and stir for 3 minutes. Add the beef stock, V-8 juice and diced tomatoes. Stir in the potatoes, carrots, corn and green beans. Add Worcestershire, hot sauce and seasonings to taste. Stir, cover and simmer over medium to medium low (low bubble) for 1-1/2 to 2 hours, or until veggies are tender, stirring occasionally. Taste and adjust seasonings as needed.

Hobo stew can be served as is, or spooned over rice or mashed potatoes. Serve with hot cornbread, biscuits or rolls.

Cook's Notes: Use other meats such as stew meat, venison, bacon, Italian sausage, smoked sausage, andouille or kielbasa - all good choices. If using a stewing type of meat, allow it to cook in the base veggies and juice for about 1 hour before adding in the vegetables. Use fresh or frozen vegetables, or may substitute 1 can of drained carrots, corn and cut green beans or use a variety of other vegetables or beans, such as mixed vegetables, shredded cabbage, black-eyed peas, pinto beans, hominy, turnips, kidney beans, great northern beans, lima beans. You'll want around 5 to 6 cups fresh or frozen, or about 3 (15 ounce) cans. If using canned vegetables, allow the potatoes and carrots to cook for 30 minutes before adding in the canned vegetables.

Also okay to use lower sodium or spicy V-8, but taste stew to adjust seasonings up or down as needed. If using spicy, take into consideration hot peppers and other spicy seasonings you use. May also substitute a can of Rotel tomatoes for the diced, if desired.

Crockpot: Saute the vegetables and brown the meat as above; add that along with the remaining ingredients to a 6 quart slow cooker, cover and cook on low for 4 to 6 hours or until veggies are tender.

Variation: To convert to a Hobo Soup, increase the beef stock/broth to about 4 cups or to desired consistency.

It's been a brisk and mostly enjoyable two weeks since the launch of Windows 8 and the start of this diary. Whereas my last entry was all about productivity, there's now been ample opportunity to relax with the new OS, play a few titles in Steam, and run some general performance benchmarks. These not-so-onerous tasks were completed using an AMD FX-based triple-monitor gaming rig upgraded from Windows 7 Ultimate to Windows 8 Pro, with all games running at 5,760 x 1,080 and max detail settings, and all the hardware was kept constant to allow a before-and-after comparison. You'll find the results after the break, along with a few broader impressions of what Windows 8 might mean for an early-adopting desktop gamer.

Wealthy Donors Increasingly Manage Giving Through Family Offices ...

A growing number of philanthropic families are looking to family offices to manage their charitable giving, according to a new study released by the National Center for Family Philanthropy.

Families are leveraging the strengths of philanthropy and the family office for greater effect in the community and more satisfaction for themselves, the report found. It also noted that both benefits and challenges attend use of the family office structure.

Family Office Exchange, a partner in the study that was sponsored by Threshold Group, has estimated that some 5,000 single-family offices operate in the U.S., and a large percentage support philanthropic activity.

Researchers surveyed family offices with charitable foundations and conducted personal interviews with some of them.

The report identified several distinct advantages for enhancing a family?s philanthropy through a family office structure:

  • Integration of functions and services
  • Economies of scale, particularly when the family office supports multiple giving vehicles or foundations
  • Efficiency for family members and external vendors
  • Expanded capacity for mission-related investments by the foundation
  • Alignment of the family?s shared values as they relate to governance, financial and investment management, and family leadership development.

?We found that managing philanthropy through the family office can be a very effective approach for families, particularly if there are multiple branches to the family and if they are using a number of charitable vehicles,? Virginia Esposito, president of the National Center, said in a statement.

The study also found that both family office leaders and foundation staff members highly valued next-generation education, though neither side spent much time in this area. The National Center recommended that both the philanthropic sector and family offices invest more focus and resources in educating and training the next generation for leadership.

Respondents also reported challenges associated with managing philanthropy through a family office structure:

  • Different cultures of business and philanthropy that can cause miscommunication and misunderstandings
  • Varying measures of success and different terminology that can lead to tension
  • Them/us mentality resulting from competing interests and priorities, and lack of respect for charitable goals and endeavors
  • Competition for the attention and resources of family members
  • Complicated and counterintuitive IRS restrictions on interactions.

?To get it right, families must pay careful attention to structure and governance, to understanding the goals and values of the family, and to effective communications,? Esposito said.

One finding surprised researchers: the extent to which family office staffers were involved in the family?s philanthropic activity. The majority of respondents said the family office was involved in helping the foundation find a strategic focus, leverage the impact of its giving and measure the effectiveness of its grants.
